포스텍 국가연구소 K-BIGHEART에서 Washington University의 김진경 교수님을 모시고 특별 초청 강연을 개최합니다.
생체 내(in vivo) 2광자 달팽이관 이미징 기술을 통해 청력 손실 없이 살아있는 생쥐의 내유모세포(IHC) 소리 유발 칼슘 반응을 세포 단위에서 직접 관측하고, 개별 유모세포가 소리의 주파수와 강도를 어떻게 부호화하는지 그 역동적인 과정을 규명한 이번 강연에 관심 있는 분들의 많은 참여를 바랍니다.

▷Abstract
The cochlea is an exceptionally sensitive and precise organ, capable of detecting sounds ranging from a ticking clock to a blasting siren. Inner hair cells (IHCs), the primary sensory cells for hearing, convert sound-evoked traveling waves along the cochlea into electrical signals that transmit auditory information to the brain. How IHCs encode the vast range of sound intensities, spanning more than six orders of magnitude, remains one of the most remarkable yet elusive features of the auditory system. While in vivo monitoring of peripheral cellular dynamics has advanced understanding across other sensory systems, achieving such high-resolution interrogation in the auditory system remains challenging due to the cochlea’s complex, fragile, fluid-filled structure and its deep location within the temporal bone. Owing to these challenges, a knowledge gap in sound encoding exists between the mechanical vibrations at the organ level and the cellular activity at the single cell/nerve level. To bridge this gap, we developed a hearing-preserved in vivo two-photon cochlear imaging method that enables direct monitoring of sound-evoked Ca2+ responses in individual IHCs in living mice. We demonstrate that GCaMP, a Ca2+ sensor designed to detect neuronal action potentials, reliably reports graded receptor potentials in IHCs. Individual IHCs exhibit sharp frequency tuning centered at their characteristic frequency (CF), with sharpness comparable to that of auditory nerve fibers measured in the same mice using forward-masked compound action potential recordings. IHC activation initiates with ~11 cells at the CF threshold and expands by recruiting ~18 additional cells with a +10 dB increase. Ca2+ response magnitudes in IHCs exhibited frequency-dependent input-output relationships across sound intensity. Most increased monotonically and linearly with intensity; however, near the CF they became non-monotonic, rising initially and then declining at higher intensities. Our in vivo data provide the first direct cellular readout of the cochlear traveling wave and reveal how individual IHCs encode sound frequency and intensity, advancing our understanding of peripheral auditory processing.
